I had this happen, but I was already starting to replace the lock actuator, but tried to fix the faltering lock actuator first. The old actuator completely failed, I hadn’t put the wires and hooks back on properly and the lock wasn’t flipping the big hook so I was screwed. Here’s what I did.

Removed door trim near handle and unscrewed the two screws (Torx 25 on a ratchet extension or a ratcheting bitdriver) that hold the inner door panel to the door. Remove the window weatherproof trim (not the soft, glued on door trim, just the stiff, velvety trim inside the door). Remove the arm rest cover and get the two screws that hold the panel to the door, one inside the top of the arm rest and one in the middle. Lift the panel out of the top of the door and bend it back as best possible without cracking it. You can get an arm’s worth plus. Much more than that and the panel cracks in two places. You can unlatch the speaker and door electronics connection and remove the big plastic door insulation the cable wire passes through to go to the inside handle. With the seat all the way back, window down, door panel peeled away at the top of the door, still screwed in at the bottom, you can reach in where you removed the insulation and pull the cables manually. If that doesn’t work there are two screws to the mechanics of the door lock actuator. You can get a stubby screwdriver or a jewelers/eyeglass repair screwdriver in there to remove those two screws. Then you have to figure out what trips the door. That’s what I did.

My new lock actuator does not communicate properly with VCDS and it doesn’t hold the door the same way the OEM did, making the passenger think their door is ajar even when the latch is closed. It has to be locked or it rattles. Booo!

    Good luck...

Thanks for the info. Quick update: I was trying to release the door using the remote - flicking lock and unlock - with no success. I then remembered the lock on the drivers door and flicked that between lock and unlock several time and hey presto! it unlocked inside and out. I have now ordered an actuator off of eBay - Front Left Door Lock Actuator Mechanism For Audi A4(B8) A5 Q3 Q5 Passenger Side 8999451115659 | eBay - and will be the following instructions on this YouTube video - How to replace the door lock actuator in an Audi A4 B8 - YouTube

Final update :all fixed now. As long as you can open the door is not very difficult to replace the actuator. Thank you to everyone who offered advice.

Good to hear that you got it resolved. Ive had the same issue with multiple doors on the A4 in the past. I have found that replacing the lock module or cleaning the motors in the module sometimes worked. When that didnt, replacing the bowden cable did work. There is more information here: Warranty Void: Repairing Audi A4 B8 8K door not unlocking with remote fob
